目的观察益气化瘀清热方及其拆方各类含药血清对脂多糖(lipopolysaccharide,LPS)诱导下体外培养的大鼠肾小球系膜细胞(primary mesangial cells,Ms C)表达Smad2及泛素mRNA的影响。方法采用RT-PCR法检测各类含药血清对Ms C表达Smad2及泛素mRNA的影响。结果各组含药血清均能抑制体外培养的Ms C表达Samd2及泛素mRNA,与对照组比较均有明显差异(P<0.01),其中复方组作用最强,优于TW组(P<0.01);清热组和化瘀组优于益气组(P<0.01),但二者比较无明显差异(P>0.05)。结论益气化瘀清热方及其拆方、TW含药血清均能抑制体外培养的大鼠Ms C表达Samd2及泛素mRNA,复方组较强,化瘀组、TW组次之,益气组较弱。
Objective To observe the effects of Yiqi Huayu Qingyu Recipe and its various components on the expression of Smad2 in primary mesangial cells (Ms C) induced by lipopolysaccharide (LPS) Effect of mRNA. Methods RT-PCR was used to detect the effect of various serum-containing drugs on Ms C expression of Smad2 and ubiquitin mRNA. Results The serum of all groups could inhibit the expression of Samd2 and ubiquitin mRNA in Ms C cells in vitro, which were significantly different from the control group (P <0.01). The compound group had the strongest effect than the TW group (P <0.01) ); Qingre and Huayu were superior to Yiqi (P <0.01), but there was no significant difference between them (P> 0.05). Conclusion Yiqi Huayu Heat Decoction and its disassembled formulas and TW containing serum can both inhibit the expression of Samd2 and ubiquitin mRNA in Ms C of rats cultured in vitro, Weaker.
