miR-199b-5p在骨肉瘤中的预测和诊断作用

目的探索miR-199b-5p在骨肉瘤中的预测和诊断作用。方法运用RT-PCR方法检测骨肉瘤患者组织中miR-199b-5p和DRAM的表达水平;利用生物信息学预测工具预测miR-199b-5p的靶基因;分析DRAM与miR-199b-5p的表达关系。结果 miR-199b-5p在组织中有表达,和癌旁正常组织相比,肿瘤组织中的miR-199b-5p表达水平显著升高(P<0.05);miR-199b-5p与患者的性别、发病年龄、病变部位、肿瘤分型和分期无明显关系(P>0.05)。研究还发现DRAM在各种组织中均有表达,而DRAM mRNA表达在两者中无明显差异,DRAM与miR-199b-5p呈负相关的表达关系。结论本研究表明miR-199b-5p可以作为骨肉瘤一个诊断标志物,而DRAM可能是miR-199b-5p的调控靶基因。 Objective To explore the predictive and diagnostic role of miR-199b-5p in osteosarcoma. Methods RT-PCR was used to detect the expression of miR-199b-5p and DRAM in the tissue of osteosarcoma patients. The bioinformatics prediction tools were used to predict the target genes of miR-199b-5p. The relationship between DRAM and miR-199b-5p expression was analyzed . Results miR-199b-5p was expressed in the tissues and the expression of miR-199b-5p in tumor tissues was significantly higher than that in normal tissues (P <0.05). The correlation between miR-199b-5p and the gender, Age, lesion, tumor type and stage had no significant relationship (P> 0.05). The study also found that DRAM was expressed in various tissues, while the expression of DRAM mRNA showed no significant difference between the two, and the expression of DRAM was negatively correlated with miR-199b-5p. Conclusion This study shows that miR-199b-5p can be used as a diagnostic marker for osteosarcoma, while DRAM may be the target of regulation of miR-199b-5p.
