目的:观察参芪复方对2型糖尿病大血管病变小鼠糖脂代谢的影响和对骨骼肌的保护作用。方法:自发性2型糖尿病KKAy小鼠60只,实验分为KKAy组、模型组、参芪复方组(14.4 mg·g-1·d-1)和罗格列酮组(1.33μg·g-1·d-1),每组15只,另设C57BL/6J小鼠15只为正常组,共5组。除正常组和KKAy组外,其余各组每日给予含一氧化氮合酶(NOS)抑制剂N-硝基-L-精氨酸甲酯(L-NAME)的饮水及高脂饲料,连续8周,建立2型糖尿病大血管病变模型。造模同时开始ig给药,期间观察动物一般状况,监测各个时间段的血糖值,实验结束后摘眼球取血,分离血清,检测总胆固醇(TC),甘油三酯(TG),高密度脂蛋白胆固醇(HDL-C)及低密度脂蛋白胆固醇(LDL-C)含量,取主动脉和腓肠肌并观察其病理形态学改变。结果:与正常组比较,KKAy组小鼠摄食量、饮水量、尿量偏多,体重较重,模型组与KKAy组小鼠血糖明显升高(P<0.01),KKAy组小鼠血糖略高于模型组,但无差异,模型组小鼠血清中TC,TG,HDL-C,LDL-C含量均明显升高(P<0.01),KKAy组小鼠血清中TC,TG,HDL-C,LDL-C含量均有所升高(P<0.05),但低于模型组小鼠,模型组腹主动脉内膜水肿,出现明显的骨骼肌细胞萎缩、水肿、断裂和炎症状况,KKAy组小鼠均也有同模型组的变化,但无模型组明显,模型组与KKAy组小鼠在体表均可见到不同程度的抓痕及伤口;与模型组比较,参芪复方可改善模型动物的一般状态,参芪复方与罗格列酮均明显降低小鼠血糖(P<0.05,P<0.01),均明显降低TC,TG,HDL-C,LDL-C含量(P<0.05,P<0.01);病理检查结果显示,参芪复方与罗格列酮均可减轻模型小鼠腹主动脉内膜水肿,减轻骨骼肌细胞萎缩、水肿、断裂和炎症状况。结论:参芪复方可改善2型糖尿病大血管病变小鼠糖脂代谢和血管损害,对骨骼肌起保护作用。
Objective: To observe the effects of Shenqi compound on glucose and lipid metabolism and protective effects on skeletal muscle of type 2 diabetic macroangiopathy mice. Methods: Sixty KKAy mice with spontaneous type 2 diabetes were divided into three groups: KKAy group, model group, Shenqi Compound group (14.4 mg · g -1 · d -1) and rosiglitazone group (1.33 μg · g -1) 1 · d-1), 15 mice in each group, and 15 mice in C57BL / 6J group as normal group. Results: Compared with the normal group, the mice in KKAy group had higher food intake, water intake and urine volume, heavier body weight, higher blood glucose in model group and KKAy group (P <0.01), and slightly higher blood glucose in KKAy group The levels of TC, TG, HDL-C and LDL-C in the model group were significantly increased (P <0.01), while the levels of TC, TG, HDL-C, (P <0.05), but lower than that of the model group, the intima of the abdominal aorta of the model group showed obvious atrophy, edema, rupture and inflammation of skeletal muscle cells, and the KKAy group was smaller Rats also have the same model group changes, but no model group was significantly, the model group and KKAy mice in the body surface can be seen varying degrees of scratches and wounds; compared with the model group, Shenqi compound can improve the general model animals (P <0.05, P <0.01), both of them significantly decreased the content of TC, TG, HDL-C and LDL-C Pathological examination showed that both Shenqi Compound and rosiglitazone could reduce the intimal edema of abdominal aorta in model mice and alleviate the atrophy, edema, rupture and inflammation of skeletal muscle cells. Conclusion: Shenqi Compound can improve blood glucose and lipid metabolism and vascular damage in type 2 diabetic rats and protect skeletal muscle.
