目的:检测凋亡相关基因在维吾尔族妇女宫颈癌中的表达,探讨维吾尔族妇女宫颈癌的发病机制及其与临床病理指标的关系。方法:收集2012年6月~2013年3月新疆维吾尔自治区人民医院妇科活检及手术切除的子宫肌瘤正常宫颈组织标本21例及宫颈癌组织标本45例,采用实时荧光定量PCR及免疫组化法对宫颈癌组织及正常宫颈组织中Bcl-x L、Bcl-2进行检测,并分析其表达水平与相应临床病理指标的关系。结果:Bcl-x L在宫颈癌组织中表达上调,与患者的肿瘤直径大小、病理分级、临床分期、肌层浸润深度及有无淋巴结转移有关。Bcl-x L mRNA在宫颈癌组织中的表达量是在正常宫颈组织中表达量的4.68倍,Bcl-2 mRNA在宫颈癌组织中的表达量低于在正常宫颈组织中的表达量。Bcl-x L、Bcl-2蛋白在宫颈癌组织和正常宫颈组织中的阳性表达率比较,差异均有统计学意义(χ2=12.357,P=0.000;χ2=5.820,P=0.015)。在宫颈癌组织中,Bcl-x L、Bcl-2蛋白在不同临床分期、肿瘤直径、临床分期、肌层浸润深度、病理分级及有无淋巴结转移中的表达水平比较,差异均有统计学意义(P<0.05)。结论:宫颈癌组织中Bcl-x L、Bcl-2的高表达可能在宫颈癌抗凋亡机制中起重要作用,这对宫颈癌的易感性预测、早期诊断、疾病监测及其分子靶向治疗的开展具有重要指导意义。
Objective: To detect the expression of apoptosis-related genes in Uigur women with cervical cancer and explore the pathogenesis of Uigur women with cervical cancer and its relationship with clinicopathological parameters. Methods: Twenty-one cases of uterine fibroids and 45 cases of cervical cancer tissues from gynecological biopsy and surgical resection of People’s Hospital of Xinjiang Uyghur Autonomous Region from June 2012 to March 2013 were collected. Real-time fluorescence quantitative PCR and immunohistochemistry The expressions of Bcl-x L and Bcl-2 in cervical cancer tissues and normal cervical tissues were detected, and the relationship between the expression of Bcl-x L and Bcl-2 and the corresponding clinicopathological parameters was analyzed. Results: The expression of Bcl-x L was up-regulated in cervical cancer, which was related to the diameter of tumor, pathological grade, clinical stage, depth of myometrial invasion and lymph node metastasis. The expression of Bcl-x L mRNA in cervical cancer tissues was 4.68 times of that in normal cervical tissues. The expression of Bcl-2 mRNA in cervical cancer tissues was lower than that in normal cervical tissues. The positive rates of Bcl-x L and Bcl-2 in cervical cancer tissues and normal cervical tissues were significantly different (χ2 = 12.357, P = 0.000; χ2 = 5.820, P = 0.015). The expression of Bcl-x L, Bcl-2 protein in cervical cancer tissues in different clinical stage, tumor diameter, clinical stage, myometrial invasion depth, pathological grading and lymph node metastasis were significantly different (P <0.05). Conclusion: The high expression of Bcl-x L and Bcl-2 in cervical cancer may play an important role in the anti-apoptotic mechanism of cervical cancer, which predict the cervical cancer susceptibility, early diagnosis, disease surveillance and molecular targeted therapy The implementation of the guide is of great significance.
