目的 :探讨 HSV- 2在体内诱导细胞凋亡的效果、形态学特点和 Fas蛋白的表达。方法 :用 HSV- 2 333株感染小鼠阴道 ,光镜下进行原位观察 ,TUNEL末端标记染色显示凋亡的细胞 ,免疫组化技术检测 Fas蛋白的表达。结果 :感染后的第 1天粘膜上皮内即出现大量的凋亡细胞 ,第 3~ 6天凋亡细胞的数量及在上皮内的分布范围达最高水平。凋亡细胞被 TUNEL标记染成棕褐色 ,经免疫组化技术证实凋亡细胞有 Fas蛋白的表达。结论 :HSV- 2 333株阴道感染可同时诱导细胞坏死及凋亡 ,Fas蛋白的表达参与了细胞凋亡的发生。细胞凋亡可能在限制病毒产生子代及限制感染区域扩展中起重要作用
Objective: To investigate the effect of HSV-2 on inducing apoptosis in vitro, the morphological characteristics and the expression of Fas protein. Methods: The vagina was infected with HSV-2 333 strain. The cells were observed under light microscope and apoptotic cells were stained with TUNEL. The expression of Fas protein was detected by immunohistochemistry. Results: A large number of apoptotic cells appeared in the mucosal epithelium on the first day after infection. The number of apoptotic cells and their distribution in the epithelium ranged from the 3rd to 6th day to the highest level. TUNEL-labeled apoptotic cells were stained tan, and the expression of Fas protein in apoptotic cells was confirmed by immunohistochemistry. Conclusion: Vaginal infection with HSV-2 333 can induce cell apoptosis and necrosis at the same time. The expression of Fas protein is involved in the apoptosis. Apoptosis may play an important role in limiting the generation of virus and limiting the expansion of the infected area
