大肠癌是我国常见消化道肿瘤,目前对大肠癌发生发展过程中的各种基因及细胞学研究已得到越来越广泛的研究。近年来,转化生长因子β-Smad信号转导通路在恶性肿瘤中的研究是肿瘤信号传导机制领域的一大热点。而该通路中的关键下游分子Smad蛋白家族更是作为一个新的靶点被广泛研究。Smad蛋白家族是由一类结构和功能相关的转录因子组成,广泛存在于多种生物的各种组织中,并在细胞增殖分化、胚胎发生、血管形成和肿瘤发生中起着重要作用,不同的Smads蛋白在大肠癌的发生发展中各自扮演不同的角色,因此,研究Smads的功能、Smads与其他细胞因子之间的相互作用及其在大肠癌中的特异性作用,对以信号通路作为靶点的大肠癌防治具有重大指导意义。
