目的:研究瓜馥木总黄酮最佳提取工艺,并将总黄酮进行富集纯化,测试其清除自由基活性。方法:采用正交实验,考察乙醇浓度、提取温度、固液比及提取时间4个因素对该工艺的影响;利用AB-8大孔吸附树脂分离纯化总黄酮粗品;采用清除DPPH自由基初步评价瓜馥木提取物及总黄酮的抗氧化能力。结果:确立了瓜馥木总黄酮的最佳提取工艺为:提取时间3 h,乙醇浓度90%,固液比1∶10,提取温度为70℃。黄酮粗品经AB-8大孔树脂纯化以后,含量可达到55%以上。结论:优选出的最佳提取工艺条件及纯化总黄酮的方法对进一步的工艺开发有较好的参考价值,瓜馥木富集纯化的总黄酮部位具有较强的清除自由基能力,为开发成为中药保健品提供了一定的依据。
Objective: To study the best extraction technology of total flavonoids from melon wood, and to enrich and purify the total flavonoids to test its scavenging free radical activity. Methods: The effects of ethanol concentration, extraction temperature, solid-liquid ratio and extraction time on the process were investigated by orthogonal experiments. The crude flavonoids were isolated and purified by AB-8 macroporous adsorption resin. The DPPH radical scavenging assay Antioxidant capacity of melon wood extract and total flavonoids. Results: The optimal extraction process of total flavonoids from melon was established as follows: the extraction time was 3 h, the ethanol concentration was 90%, the solid-liquid ratio was 1:10 and the extraction temperature was 70 ℃. After the crude flavonoids are purified by AB-8 macroporous resin, the content can reach more than 55%. Conclusion: The optimal extraction conditions and the method of purification of total flavonoids have good reference value for further process development. The enrichment and purification of total flavonoids from Fructus Forsythiae has a strong ability to scavenge free radicals, Chinese health products provide some basis.
