用记录搏动频率、细胞化学及生物化学方法,研究了镉对体外培养乳大鼠心肌细胞的毒作用。结果发现,培养的心肌细胞停搏率与镉剂量呈直线正相关,CdCl_2的半数停搏浓度为4.01μg/ml。细胞内糖原、乳酸脱氢酶(LDH)及琥珀酸脱氢酶(SDH)含量分别与镉剂量呈负相关关系。培养液LDH活力在给毒前后无显著差异,表明细胞尚未崩溃。以上结果提示,镉严重损害细胞能量代谢,导致心肌细胞停搏。停搏是镉中毒的潜在性损害。
The toxic effects of cadmium on cardiomyocytes cultured in vitro were studied by recording pulsatile frequency, cytochemical and biochemical methods. The results showed that there was a linear positive correlation between the rate of cardiomyocyte cardiomyocyte arrest and cadmium dose, and the half number of CdCl 2 was 4.01 μg / ml. Intracellular glycogen, lactate dehydrogenase (LDH) and succinate dehydrogenase (SDH) content were negatively correlated with the amount of cadmium. LDH activity in the culture medium before and after giving no significant difference, indicating that cells have not collapsed. The above results suggest that cadmium seriously damages the energy metabolism of the cells and causes cardiomyocyte arrest. Cardiac arrest is a potential damage to cadmium poisoning.
