目的:观察加味五子衍宗颗粒及其有效组分对β-淀粉样肽所致老年性痴呆(AD)模型鼠空间学习记忆能力的影响。方法:采用大鼠侧脑室立体定向注射Aβ25-35的方法建立AD动物模型,服用加味五子衍宗颗粒及其总黄酮、总多糖15d后以全自动Morris水迷宫实验检测大鼠定位航行试验及空间探索试验,来评价大鼠空间学习记忆功能。结果:加味五子衍宗颗粒及其有效组分总黄酮、总多糖均能明显缩短模型大鼠在Morris水迷宫定位航行试验中的逃避潜伏期和搜索距离(P<0.05),显著增加空间探索试验中原平台所在象限搜索时间和距离占总时间和总距离的百分比(P<0.05,P<0.01)。结论:加味五子衍宗颗粒及其总黄酮、总多糖能改善由β-淀粉样肽所致AD模型鼠的空间学习记忆能力。
Objective: To observe the effects of Jiawei Wuzi Yanzong Granule and its effective components on the spatial learning and memory of Alzheimer ’s disease (AD) model rats induced by β - amyloid peptide. Methods: Animal models of AD were established by injection of Aβ25-35 into the lateral ventricle of rats. The rats were treated with Modified WZYR and its total flavonoids and total polysaccharide for 15 days. Explore the test to evaluate the spatial learning and memory function of rats. Results: Modified Wuzi Yanzong granule and its active ingredient total flavonoids and total polysaccharides significantly shortened the escape latency and search distance (P <0.05) of the model rats in the Morris water maze positioning navigation test, significantly increased the spatial distribution of Zhongyuan The percentage of search time and distance of the platform where the platform was located in total time and total distance (P <0.05, P <0.01). Conclusion: Jiawei Wuzi Yanzong Granule and its total flavonoids and total polysaccharides can improve spatial learning and memory of AD model rats induced by β-amyloid peptide.
