目的 研制对神经传导递质多巴胺 (DA)具有高灵敏度高选择性的微型生物传感器 ,用于活体大鼠的铅神经毒理学研究。方法 用制备的微型生物传感器 ,用电化学分析方法研究活体大鼠经铅染毒后神经递质多巴胺的动态变化。结果 研制的微型生物传感器对DA的检出下限为 32 .5nmol/L ,灵敏度高 ,选择性好 ,在脑内可以连续工作 10h以上。应用该传感器观察了铅染毒后活体大鼠脑纹状体内神经递质多巴胺的动态变化 ,首次得到了 1/2LD50 铅经腹腔染毒后活体大鼠神经递质多巴胺的时间效应动态变化曲线。结论 该微型生物传感器可用于脑内神经递质多巴胺的活体研究。
Objective To develop a miniature biosensor with high sensitivity and selectivity to neurotransmitter dopamine (DA) for lead neurotoxicology in living rats. Methods Using the prepared micro-biosensor, the dynamic changes of neurotransmitter dopamine in living rats after lead exposure were studied by electrochemical analysis. Results The miniature biosensor developed for DA detection limit of 32.5nmol / L, high sensitivity, good selectivity, continuous work in the brain more than 10h. The sensor was used to observe the dynamic changes of neurotransmitter dopamine in the striatum of live rats after lead exposure. The dynamic change of the time effect of dopamine, a neurotransmitter in living rats after 1 / 2LD50 lead exposure for the first time, was obtained. Conclusion The miniature biosensor can be used for the in vivo study of neurotransmitter dopamine in the brain.
