目的探讨肝X受体β(liver X receptorβ,LXRβ)在皮肤鳞状细胞癌(cutaneous squamous cell carcinoma,cSCC)中的表达及其意义。方法采用PCR array、Western blot检测LXRs在角质形成细胞(keratinocyte,KC)和皮肤鳞状细胞癌A431细胞系中的表达,并采用免疫组化Envision法检测LXRβ在52例日光性角化病(actinic keratosis,AK)、40例鲍温病(Bowen’s disease,BD)、60例cSCC中的表达,分析LXRβ表达与患者临床参数之间的关系。结果与KC比较,A431细胞中LXRα、LXRβ的mRNA和蛋白水平均升高,以LXRβ升高尤为显著。免疫组化染色结果发现LXRβ在AK、BD、cSCC中表达逐渐升高,以cSCC升高最明显,其表达与肿瘤分化程度、瘤体大小、浸润深度相关(P<0.05),与年龄、性别、发病部位无关(P>0.05)。结论 LXRβ在鳞状细胞癌细胞和组织中表达均升高,且与肿瘤的增殖、分化相关。
Objective To investigate the expression of liver X receptor β (LXRβ) in cutaneous squamous cell carcinoma (cSCC) and its significance. Methods The expression of LXRs in keratinocyte (KC) and cutaneous squamous cell carcinoma A431 cell lines was detected by PCR array and Western blot. Immunohistochemical Envision method was used to detect the expression of LXRβ in 52 cases of actinic keratosis, AK), 40 cases of Bowen’s disease (BD) and 60 cases of cSCC. The relationship between the expression of LXRβ and the clinical parameters was analyzed. Results Compared with KC, the mRNA and protein levels of LXRα and LXRβ were increased in A431 cells, especially in LXRβ. The results of immunohistochemistry showed that the expression of LXRβ in AK, BD and cSCC increased gradually, and the expression of LXRβ was the most obvious in cSCC. The expression of LXRβ was correlated with tumor differentiation, tumor size and depth of invasion (P <0.05) , The incidence of site has nothing to do (P> 0.05). Conclusion The expression of LXRβ in squamous cell carcinoma tissues and tissues are increased, and it is related to tumor proliferation and differentiation.
