目的 分析日本血吸虫成虫呕吐物与感染血清的免疫反应性 ,寻找特异性的血吸虫病诊断抗原。方法 采用 SDS- PAGE和免疫印迹分析呕吐物的免疫反应性 ,过碘酸钠氧化处理以鉴定呕吐物的抗原决定簇的性质。结果 日本血吸虫成虫呕吐物中存在能被血吸虫感染血清识别的特异抗原 31/ 32、4 7、6 0 / 6 2 k Da蛋白及 6 7k Da蛋白 ;31/ 32 k Da蛋白和 6 7k Da蛋白的抗原表位为蛋白 ,而 4 7k Da蛋白和 6 0 / 6 2 k Da蛋白的抗原表位为糖基。结论 日本血吸虫成虫呕吐物中存在血吸虫感染血清识别的特异抗原 ,其中 31/ 32 k Da蛋白已报告可用于血吸虫病诊断 ,4 7k Da蛋白和 6 0 / 6 2k Da蛋白含糖基表位并具有潜在的诊断价值。
Objective To analyze the immunological reactivity of adult vomitus and infected sera of Schistosoma japonicum to find out the specific diagnostic antigen of schistosomiasis. Methods The immunoreactivity of vomit was analyzed by SDS-PAGE and Western blotting, and the oxidation of sodium periodate was performed to identify the antigenic determinants of vomit. Results There were 31/32, 47, 60/62 kDa proteins and 67 kDa proteins recognized by schistosome-infected sera in the adult vomit of Schistosoma japonicum; 31/32 kDa protein and 67 kDa protein The antigenic epitope is a protein, whereas the 4 7kDa protein and the 6 0/62 2 kDa antigen epitope are glycosylated. Conclusion The specific antigen recognized by schistosome-infected sera is present in the adult vomitus of Schistosoma japonicum. The 31/32 kDa protein has been reported to be useful in the diagnosis of schistosomiasis, the 47kDa protein and the 60kDa protein glycosylation epitope and has Potential diagnostic value.
