目的:探讨黑蒜提取液(ABGE)对荷瘤小鼠Panc-1细胞在体内的生长以及转移能力的影响,并初步探讨其分子机制。方法:分别建立胰腺癌Panc-1小鼠皮下移植瘤模型以及转移瘤模型,将40只小鼠随机分为空白对照组,黑蒜低,高剂量组、阳性对照组4组,分别给予0.9%氯化钠溶液,黑蒜提取液组0.4、0.8m L(相当于生药8.0、16.0g/kg),阳性环磷酰胺,2周后摘取眼球血脱臼处死小鼠,剥离瘤体称重,计算肿瘤生长抑制率以及转移抑制率;免疫组化检测肿瘤组织Smad4以及TGF-β1的表达。结果:腹腔注射黑蒜提取液可显著抑制小鼠Panc-1细胞移植瘤生长及肺转移的发生率,肿瘤抑制率可高达63.6%(P<0.01),而肺转移抑制率可高达63.5%(P<0.01)。且黑蒜高剂量抑制Panc-1细胞转移作用优于阳性对照组;免疫组化可检测发现:黑蒜提取液组Panc-1细胞胞质中TGF-β1褐黄色颗粒较空白对照组少见,而Smad4胞浆着黄色则较空白对照组多见。结论:腹腔注射黑蒜提取液可显著抑制小鼠Panc-1细胞移植瘤生长及肺转移的发生率,上调荷Panc-1小鼠移植瘤中Smad4的表达并下调TGF-β1的表达,与TGF-β1/Smad4信号通路密切相关,以上可能是黑蒜提取液抑制胰腺癌Panc-1细胞生长和转移的重要作用机制。
Objective: To investigate the effect of ABGE on the growth and metastasis of Panc-1 cells in vivo in mice and to investigate its molecular mechanism. Methods: Panc-1 pancreatic tumor model and metastatic tumor model were established respectively. The tumor inhibition rate was as high as 63.6% (P <0.01), while the lung metastasis rate was as high as 63.5% P <0.01). The high dose of black garlic could inhibit the metastasis of Panc-1 cells better than the positive control group. Immunohistochemistry showed that TGF-β1 brown granules in the cytoplasm of Panc-1 cells were rare in the black garlic extract group compared with the blank control group Smad4 cytoplasm yellow than the blank control group more common. Conclusion: The intraperitoneal injection of black garlic extract can significantly inhibit the growth of Panc-1 cells in mice and the incidence of lung metastases, upregulate the expression of Smad4 and down-regulate the expression of Smad4 in Panc-1 mice xenografts, -β1 / Smad4 signaling pathway is closely related to the above may be black garlic extract Panc-1 cells inhibit pancreatic cancer cell growth and metastasis of the important mechanism of action.
