目的:研究甘蔗渣中多糖的最佳纯化工艺。方法:用Savage法、TCA法脱蛋白;用双氧水法、活性炭法脱色;用AB-8,DB301,D101,MG-1大孔树脂纯化甘蔗渣多糖,用苯酚-硫酸法测定多糖的含量。结果:最佳工艺为用Savage法脱蛋白、活性炭脱色、MG-1型大孔吸附树脂纯化(上样量为0.3 g,柱高35 cm、流速10 mL/15 min),经富集后甘蔗渣多糖的含量由51.11%提高到了83.57%。结论:本实验为甘蔗渣多糖的分离纯化条件提供了参考,为甘蔗渣的进一步开发研究提供了依据。
Objective: To study the best purification technology of polysaccharide in bagasse. Methods: Deproteinization was performed by Savage method and TCA method. Decolorization was performed by hydrogen peroxide method and activated carbon method. Polysaccharide was purified with AB-8, DB301, D101 and MG-1 macroporous resins. Polysaccharide content was determined by phenol-sulfuric acid method. Results: The optimum conditions were as follows: the protein was deproteinized by Savage method, the activated carbon was decolorized, and the MG-1 macroporous resin was purified (loading amount was 0.3 g, column height 35 cm and flow rate 10 mL / 15 min) The content of polysaccharides increased from 51.11% to 83.57%. Conclusion: This experiment provides a reference for the isolation and purification of sugarcane bagasse polysaccharides, and provides the basis for the further development of sugarcane bagasse.
