目的探讨胃炎饮对实验性胆汁反流性胃炎大鼠胃黏膜环氧合酶-2(COX-2)mRNA表达的影响。方法采用自制反流液灌胃建立实验性胆汁反流性胃炎大鼠模型,以吗叮啉为对照,采用RT-PCR技术观察胃炎饮对实验性胆汁反流性胃炎大鼠胃黏膜COX-2mRNA表达以及病理组织学的影响。结果模型组大鼠胃黏膜出现明显破损、脱落,黏膜下可见大量炎细胞浸润,大部分大鼠可见不典型增生和肠上皮化生,同时胃黏膜COX-2mRNA表达升高(P<0.01),与模型组比较,胃炎饮可明显改善实验性胆汁反流性胃炎大鼠胃黏膜病理改变,并降低COX-2mRNA表达(P<0.05或P<0.01)。结论胃炎饮具有降低模型大鼠胃黏膜COX-2mRNA表达,防止胃黏膜肠上皮化生、不典型增生等癌前病变发生的作用。
Objective To investigate the effect of Weiyanyin on the expression of cyclooxygenase-2 (COX-2) mRNA in gastric mucosa of rats with experimental bile reflux gastritis. Methods A rat model of experimental bile reflux gastritis was established by intragastric administration of self-made reflux fluid. The effect of Weiyan Yin on COX-2 mRNA in gastric mucosa of rats with experimental bile reflux gastritis was observed by RT-PCR. Expression and histopathology. Results The gastric mucosa of rats in the model group was significantly damaged and detached. Substantial inflammatory cells infiltrated under the mucosa. Most of the rats showed dysplasia and intestinal metaplasia, and the expression of COX-2 mRNA in gastric mucosa was increased (P<0.01). Compared with the model group, Weiyan Yin can significantly improve the pathological changes of gastric mucosa in rats with experimental bile reflux gastritis, and reduce COX-2 mRNA expression (P<0.05 or P<0.01). Conclusion Weiyanyin can reduce the expression of COX-2 mRNA in gastric mucosa of model rats and prevent the occurrence of precancerous lesions such as gastric mucosal intestinal metaplasia and atypical hyperplasia.
