目的探讨miR-630对胃癌细胞HGC-27增殖、凋亡能力的影响,发现miR-630在胃癌发生发展中所发挥的作用,寻找胃癌发生背后的分子机理和机制,帮助找到新的胃癌临床诊断标志物,甚至开发新的临床治疗方法。方法使用脂质体将miR-630 mimics和inhibitor转染HGC-27细胞,同时以mimics control、inhibitor control作为阴性对照。采用细胞增殖实验观察miR-630对胃癌细胞HGC-27增殖能力的影响。采用流式细胞术检测miR-630对胃癌细胞HGC-27凋亡情况的影响。结果细胞增殖实验显示,miR-630 mimics可以抑制胃癌细胞HGC-27的增殖能力,而inhibitors可以促进胃癌细胞HGC-27的增殖能力;流式细胞术结果显示,miR-630 mimics可以促进胃癌细胞HGC-27的凋亡,而inhibitors则抑制胃癌细胞HGC-27的凋亡,说明miR-630不仅抑制胃癌细胞HGC-27的增殖同时促进其凋亡,实验结果差异均有统计学意义(P<0.05)。结论 miR-630能通过促进胃癌细胞HGC-27的凋亡抑制胃癌细胞增殖,可能在胃癌的发生和进展中发挥重要作用。
Objective To investigate the effect of miR-630 on the proliferation and apoptosis of gastric cancer cell line HGC-27 and to find out the role of miR-630 in the development of gastric cancer and to find out the molecular mechanism and mechanism behind the occurrence of gastric cancer and to find a new clinical diagnosis of gastric cancer Markers, and even develop new clinical treatments. Methods Liposomes were used to transfect miR-630 mimics and inhibitor into HGC-27 cells. Meanwhile, mimics control and inhibitor control were used as negative controls. Effect of miR-630 on the proliferation of gastric cancer cell HGC-27 was observed by cell proliferation assay. The effect of miR-630 on the apoptosis of gastric cancer cell HGC-27 was detected by flow cytometry. The results of cell proliferation experiments showed that miR-630 mimics can inhibit the proliferation of gastric cancer cells HGC-27, and inhibitors can promote the proliferation of gastric cancer cells HGC-27; flow cytometry results show that miR-630 mimics can promote gastric cancer cells HGC -27, while inhibitors inhibited the apoptosis of gastric cancer cell HGC-27, indicating that miR-630 not only inhibits the proliferation of gastric cancer cell HGC-27, but also promotes the apoptosis of HGC-27 cells. The difference of the experimental results was statistically significant (P <0.05 ). Conclusion miR-630 can inhibit the proliferation of gastric cancer cells by promoting the apoptosis of gastric cancer cell HGC-27, which may play an important role in the occurrence and progression of gastric cancer.
