目的探讨膜联蛋白A7表达减少是否对人宫颈癌HeLa细胞系的增殖和凋亡产生影响。方法采用Western blotting鉴定siRNA可否有效抑制膜联蛋白A7表达,然后用脂质体转染法将siRNA转染入HeLa细胞,将细胞分为siRNA干扰组、阴性对照组和空白对照组,分别在转染后48h进行刃天青实验以观察细胞增殖状况的改变;流式细胞术检测3组细胞凋亡情况。结果在转染膜联蛋白A7的siRNA后48h的HeLa细胞,刃天青实验可见siRNA干扰组细胞增殖较阴性对照组和空白对照组显著降低(P<0.05);流式细胞术实验可见siRNA干扰组细胞凋亡显著增多(P<0.05)。结论膜联蛋白A7的低表达可能影响HeLa细胞的增殖和凋亡。
Objective To investigate whether the decrease of annexin A7 expression affects the proliferation and apoptosis of human cervical cancer HeLa cell line. Methods Western blotting was used to identify whether siRNA can effectively inhibit the expression of Annexin A7. Then, siRNA was transfected into HeLa cells by lipofection method. The cells were divided into siRNA interference group, negative control group and blank control group. 48 hours after dyeing, resazurin experiments were performed to observe the changes of cell proliferation status. Flow cytometry was used to detect the apoptosis of the three groups. Results HeLa cells and resazurin transfected HeLa cells transfected with annexin A7 siRNA at 48h showed that the proliferation of siRNA-siRNA group was significantly lower than that of negative control group and blank control group (P <0.05). The results of flow cytometry showed that siRNA interference Group apoptosis increased significantly (P <0.05). Conclusion The low expression of Annexin A7 may affect the proliferation and apoptosis of HeLa cells.
