目的观察热休克蛋白27(HSP27)在砷致Ha Ca T恶性转化细胞中的水平变化情况,探讨其在砷致Ha Ca T细胞恶性转变中的作用。方法用三氧化二砷作为诱导剂,建立Ha Ca T细胞恶性转化模型,用RT-PCR方法检测HSP27mRNA的表达情况,用Western blot法检测HSP27蛋白水平。结果 Ha Ca T恶性转化细胞HSP27mRNA表达和蛋白水平均较对照组上调,差异均有统计学意义(P﹤0.05)。结论Ha Ca T细胞长期在含三氧化二砷培养基中培养可使HSP27mRNA表达和蛋白水平均上调,提示HSP27在砷诱导Ha Ca T细胞恶性转变过程中发挥着一定的作用。
Objective To observe the changes of heat shock protein 27 (HSP27) in malignant transformation of HaCa T cells induced by arsenic and to explore its role in the malignant transformation of HaCa T cells induced by arsenic. Methods Arsenic trioxide was used as an inducer to establish a malignant transformation model of HaCa T cells. The expression of HSP27 mRNA was detected by RT-PCR and the protein level of HSP27 was detected by Western blot. Results The expression of HSP27 mRNA and protein in Ha Ca T malignant transformed cells were significantly up-regulated compared with the control group (P <0.05). CONCLUSION: Long-term treatment of HaCa T cells with arsenic trioxide can up-regulate the expression of HSP27 mRNA and protein, suggesting that HSP27 may play a role in the progression of Arsenic-induced HaCa T cell malignant transformation.
