目的研究某市2014年公共场所集中空调系统水中嗜肺军团菌的分子指纹图谱,初步建立某市嗜肺军团菌的脉冲场凝胶电泳(PFGE)分型数据库。方法对某市2014年公共场所集中空调系统水分离的34株嗜肺军团菌,采用PFGE技术进行电泳,并经Bio Numerics 6.6软件进行聚类分析。结果34株嗜肺军团菌的PFGE图谱共分为24种PFGE带型,不同地区嗜肺军团菌存在同样的酶切图谱,同一采样场所不同冷却塔检出菌株的PFGE带型相同,聚类分析结果显示菌株间相似性系数为23.80%~100.00%。结论某市2014年公共场所集中空调系统水嗜肺军团菌菌株基因型散在分布,呈多态性和复杂性,同时具有克隆化特征。
Objective To study the molecular fingerprinting of Legionella pneumophila in water of central air-conditioning system in public places in a city in 2014 and to establish the PFGE typing database of Legionella pneumophila in a certain city. Methods 34 strains of Legionella pneumophila isolated from the water of centralized air-conditioning system in public places in a city in 2014 were electrophoresed by PFGE and clustered by Bio Numerics 6.6 software. Results The PFGE profiles of 34 Legionella pneumophila strains were divided into 24 PFGE patterns. The same restriction map of Legionella pneumophila existed in different regions. The PFGE patterns of the strains detected by different cooling towers in the same sampling site were the same. Cluster analysis The results showed that the similarity coefficient between strains was 23.80% ~ 100.00%. Conclusion In 2014, the genotypes of Legionella pneumophila in central air-conditioning system of public places in a city were scattered, showing polymorphism and complexity, and cloned.
