目的探讨花色苷矢车菊-3-葡萄糖苷(Cy-3g)对健康人血小板活化和聚集的影响及其可能的机制。方法流式细胞仪测定全血中血小板活化膜糖蛋白IIbIIIa(GPIIbIIIa)和P-选择素(P-selectin)的阳性表达率;血小板聚集仪检测腺苷二磷酸(ADP)诱导的最大聚集率;荧光酶标仪分析Cy-3g对血小板胞浆钙离子水平的影响。结果 50μmol/L Cy-3g组GP IIb IIIa表达率[(73.92±14.02)%]与对照组[(93.36±2.02)%]比较,差异有统计学意义(P<0.05);P-选择素的表达率各组间差异无统计学意义(P>0.05);不同浓度的Cy-3g均在体外明显抑制富集血小板血浆的聚集(P<0.001);50μmol/L Cy-3g组可以明显抑制ADP诱导的血小板钙离子荧光强度值达(51.73±6.89)%(P<0.01)。结论花色苷Cy-3g可以抑制由ADP诱导的血小板活化GPIIbIIa的表达和血小板体外聚集,血小板胞浆钙离子浓度的降低可能是花色苷抑制作用的机制之一,提示Cy-3g对于预防心血管疾病具有积极作用。
Objective To investigate the effect of cyanidin-3-glucoside (Cy-3g) on platelet activation and aggregation in healthy volunteers and its possible mechanism. Methods The positive rate of GPIIbIIIa and P-selectin in whole blood was determined by flow cytometry. The maximum concentration of adenosine diphosphate (ADP) induced by platelet aggregation was detected by platelet aggregation assay. Effect of Cy-3g on platelet cytoplasmic Ca2 + level by fluorescence microplate reader. Results Compared with the control group [(93.36 ± 2.02)%], the expression of GP IIb IIIa in 50μmol / L Cy-3g group was significantly higher than that in the control group [(73.92 ± 14.02)%] (P <0.05) (P> 0.05). Cy-3g at different concentrations significantly inhibited the accumulation of platelet-rich plasma in vitro (P <0.001), while 50μmol / L Cy-3g significantly inhibited ADP The induced platelet calcium fluorescence intensity reached (51.73 ± 6.89)% (P <0.01). Conclusions Anthocyanin Cy-3g can inhibit the expression of platelet-activating GPIIbIIa induced by ADP and platelet aggregation in vitro. The decrease of platelet cytoplasmic calcium ion concentration may be one of the mechanisms of anthocyanin inhibition, suggesting that Cy-3g can prevent cardiovascular diseases Has a positive effect.
