目的:探讨白细胞介素-1β(IL-1β)在体外培养的人早孕期细胞滋养细胞对NFκ-B的激活作用。方法:体外无血清培养的人早孕期细胞滋养细胞,以IL-1β(20μg/L)作为处理因素,通过Western blot法检测IκBα的蛋白水平。结果:L-1β作用5 min即可见IκBα的水平下降,但与作用前比较无显著性差异,15 min时IκBα的水平显著降低(P<0.01),30 min时不能检测出来;随后IκBα的水平渐渐回升,60 min左右接近未处理时的水平。结论:早孕期存在于母体和胎儿界面的IL-1β可能通过自分泌或旁分泌的方式引起NF-κB的短暂激活。
AIM: To investigate the effect of interleukin-1β (IL-1β) on the activation of NFκ-B by human cytotrophoblasts cultured in vitro in early pregnancy. Methods: Human cytokine-stimulated cells in early pregnancy were cultured in serum-free medium. IL-1β (20μg / L) was used as the treatment factor to detect the protein level of IκBα by Western blot. Results: The level of IκBα decreased after treated with L-1β for 5 min, but there was no significant difference compared with that before treatment. The level of IκBα was significantly decreased at 15 min (P <0.01), and could not be detected at 30 min. The level of IκBα Gradually pick up, 60 min or so close to the untreated level. CONCLUSION: IL-1β, which exists in the maternal and fetal interface in early pregnancy, may cause transient activation of NF-κB by autocrine or paracrine manner.
