目的 :观察局部点眼后抗真菌药特比萘芬 (terbinafine)在兔角膜及房水中的浓度。方法 :用 0 2 %特比萘芬眼液(无菌蓖麻油新鲜配置 )给兔点眼两次 ,间隔 5分钟 ,总量约 40微升。于两次点药后 5、15、30、6 0、12 0及 2 40分钟处死动物 ,取角膜及房水 ,用高效液相层析法测定角膜及房水中特比萘芬的浓度。结果 :点眼后角膜中的特比萘芬浓度逐渐增高 ,在 30分钟后达到峰值 1 39μg/g ,之后不断下降 ,点药后 2 40分钟时 ,浓度降为 0 18μg/g ;房水中特比萘芬浓度在点眼后 15分钟达到最高 ,为 5 6 2ng/ml,点药 6 0分钟之后 ,房水中特比萘芬浓度下降明显 ,到 12 0分钟时高效液相层析法未测到特比萘芬。结论 :0 2 %特比萘芬滴眼可以渗透到角膜及房水中 ,并达到有效抑菌浓度
OBJECTIVE: To observe the concentration of terbinafine, an antifungal agent, in the cornea and aqueous humor of rabbits after local point eye surgery. METHODS: Rabbits were spotted twice with 0 2% terbinafine eye drops (sterile castor oil freshly placed) for 5 minutes at a total of about 40 μl. Animals were sacrificed at 5, 15, 30, 60, 120, and 240 minutes after two drug injections. Corneas and aqueous humor were obtained. The concentration of terbinafine in corneal fluid and aqueous humor was determined by high performance liquid chromatography. Results: The concentration of terbinafine in the cornea gradually increased after reaching the peak value of 399μg / g after 30 minutes, and then decreased continuously. The concentration decreased to 0 18μg / g at 240 minutes after the point injection; The concentration of naphthalene reached the highest point 15 minutes after the point eye, which was 562ng / ml. After 60 minutes, the concentration of terbinafine in aqueous humor decreased significantly, and no effect was found by HPLC at 120 minutes Bifenafen. Conclusion: 0 2% terbinafine eye drops can penetrate the cornea and aqueous humor, and achieve effective inhibitory concentration
