目的检测巨细胞病毒CMV是否能够感染并影响小鼠腹腔巨噬细胞。方法以小鼠巨细胞病毒MCMV攻击小鼠腹腔巨噬细胞后,用地高辛标记的152bpMCMVDNA探针进行原位杂交测定。结合受染细胞形态学改变,巨细胞包涵体检测,以及细胞表面Fc受体表达Fc花环形成的变化,观察MCMV对单核-巨噬细胞的作用。结果原位杂交技术检测表明受MCMV攻击的巨噬细胞的胞浆和核内,有杂交阳性的灰蓝色颗粒沉积,与阳性对照细胞片小鼠成纤维细胞3T3相一致;未受MCMV攻击的巨噬细胞片阴性对照中,则未见有着色颗粒。同时发现,受染细胞的形态改变,核呈现典型的嗜酸性包涵体;受染细胞表面Fc受体表达呈上调趋势。结论巨细胞病毒能够感染巨噬细胞并影响其形态和功能。
Objective To detect whether cytomegalovirus CMV can infect and affect murine peritoneal macrophages. Methods Mouse peritoneal macrophages were challenged with mouse cytomegalovirus (MCMV), and then detected by in situ hybridization with digoxigenin labeled 152bp MVDNA probe. Combined with morphological changes of infected cells, detection of giant cell inclusion bodies, and changes of Fc rosette formation on the cell surface Fc receptor expression, the effect of MCMV on monocyte-macrophage cells was observed. Results In situ hybridization assay showed that MCMV-challenged macrophages had cytoplasm and nuclei with hybrid-positive gray-blue granules that were consistent with the positive control cells in 3T3 fibroblast-derived mouse fibroblasts. The non-MCMV-challenged In macrophage negative control, no colored particles were seen. At the same time, it was found that the morphology of the infected cells changed, and the nucleus showed the typical eosinophilic inclusion bodies. The expression of Fc receptors on the infected cells showed an upward trend. Conclusion Cytomegalovirus can infect macrophages and affect its morphology and function.
