目的观察扶正解毒方对肺炎链球菌感染大鼠炎性细胞因子及免疫指标的影响,探讨扶正解毒法对肺感染后免疫失衡的调控机制。方法气管内注射肺炎链球菌建立肺炎感染模型,分别用扶正解毒中药、头孢呋辛酯、扶正解毒中药+头孢呋辛酯干预,分别于给药第4日和第7日取材。观察各组肺组织形态学,ELISA检测血清及肺组织中肿瘤坏死因子-α(TNF-α)、白介素-17(IL-17)的表达;West blot检测脾组织中CD4+、CD8+水平。结果与正常组相比,模型组肺组织与血清中TNF-α、IL-17含量增高(P<0.05)。各治疗组TNF-α、IL-17水平降低,CD4+、CD8+水平增高,免疫紊乱状态改善。结论扶正解毒中药可以降低炎性细胞因子水平、增强免疫功能,改善动物生存状态。扶正解毒中药对肺炎的治疗机制可能与降低炎性细胞因子水平,减轻炎性反应,增强免疫功能有关。
Objective To observe the effect of Fuzheng Jiedu Fang on inflammatory cytokines and immune indexes in rats infected with Streptococcus pneumoniae and to explore the regulatory mechanism of Fuzheng detoxification on immune imbalance after lung infection. Methods Streptococcus pneumoniae was injected intratracheally to establish a model of pneumonia infection. Fuzheng detoxification traditional Chinese medicine, cefuroxime axetil, Fuzheng detoxification traditional Chinese medicine and cefuroxime axetil were respectively used to make the model of pneumonia infection. The lung histomorphology of each group was observed. The levels of TNF-α and IL-17 in serum and lung tissue were measured by ELISA. The levels of CD4 + and CD8 + in the spleen were detected by Western blot. Results Compared with the normal group, the content of TNF-α and IL-17 in the lung tissue and serum in the model group increased (P <0.05). The levels of TNF-α and IL-17 in each treatment group decreased, the levels of CD4 + and CD8 + increased and the immune disorders improved. Conclusion Fuzheng detoxification traditional Chinese medicine can reduce the level of inflammatory cytokines, enhance immune function and improve the living conditions of animals. Fuzheng detoxification traditional Chinese medicine treatment of pneumonia may be related to the mechanism of reducing the level of inflammatory cytokines, reducing inflammatory reactions and enhancing immune function.
