探讨辐射损伤对粘膜免疫的影响。方法小鼠全身照射8Gy后,分离培养小肠上皮内淋巴细胞,并对其数量、细胞增殖活力、细胞毒力、TNF-α和TGF-β分泌水平进行检测。结果小肠上皮内淋巴细胞数量明显下降,增殖活力和细胞毒力明显受抑,都表现为伤后8小时下降,72小时达最低值。上皮内淋巴细胞分泌TNF-α和TGF-β显著增多,表现为伤后8小时增高,72小时达峰值。结论小肠上皮内淋巴细胞数量减少和功能降低成为全身放射损伤时造成肠粘膜免疫屏障功能受损的一个重要原因。
To investigate the impact of radiation damage on mucosal immunity. Methods After 8Gy whole body irradiation, the intra-intestinal epithelial lymphocytes were isolated and cultured, and the quantity, cell proliferation activity, cytotoxicity, TNF-α and TGF-β secretion were detected. Results The number of lymphocytes in the small intestine was significantly decreased, and the proliferative activity and cytotoxicity were significantly inhibited. Both of them showed a decrease of 8 hours after injury and a minimum of 72 hours. Intraepithelial lymphocytes secreted TNF-α and TGF-β significantly increased, manifested as increased 8 hours after injury, peaked at 72 hours. Conclusion The decrease of intestinal lymphocytes and the decrease of the function of intestinal epithelial cells are the important reasons for the impaired immune barrier function of intestinal mucosa during systemic radiation injury.
