目的探讨热休克蛋白90(HSP90)的两个亚型HSP90α和HSP90β及抑癌基因p53蛋白在涎腺良性、恶性肿瘤中的表达特征及相关性。方法采用免疫组化SP法检测41例良性和17例恶性肿瘤中HSP90α、HSP90β和p53的表达。结果 (1)HSP90α和HSP90β阳性以肿瘤上皮的胞浆表达为主,伴少量胞核表达;p53表达于肿瘤上皮的胞核,伴少量胞浆表达。(2)HSP90α和HSP90β在涎腺肿瘤中均明显表达,且表达强度正相关(P<0.01),尤其HSP90α在恶性肿瘤中的阳性表达率显著高于良性肿瘤(P<0.05)。(3)p53在肿瘤中高表达为突变型,p53在恶性肿瘤的表达显著高于良性肿瘤(P<0.05)。(4)在HSP90α和HSP90β阳性时,恶性肿瘤p53的表达率均显著高于良性肿瘤(P<0.01),而且p53阳性病例HSP90α的表达率在恶性肿瘤也显著高于良性肿瘤(P<0.05)。结论 (1)HSP90α和HSP90β、突变p53的持续刺激均是涎腺恶性肿瘤发生发展的基础。(2)HSP90α和HSP90β均可用作涎腺肿瘤治疗的重要的靶点。(3)HSP90α可能通过突变的p53蛋白促进肿瘤生长及恶性转化。
Objective To investigate the expression and correlation of two subtypes of HSP90α, HSP90β and tumor suppressor gene p53 in salivary gland benign and malignant tumors. Methods Immunohistochemical SP method was used to detect the expression of HSP90α, HSP90β and p53 in 41 cases of benign and 17 cases of malignant tumors. Results (1) HSP90α and HSP90β were mainly expressed in the cytoplasm of tumor epithelium with a small amount of nucleus. P53 was expressed in the nucleus of tumor epithelium with a small amount of cytoplasm. (2) The expression of HSP90α and HSP90β in salivary gland tumors was significantly correlated with the expression intensity (P <0.01). Especially, the positive rate of HSP90α in malignant tumors was significantly higher than that in benign tumors (P <0.05). (3) p53 is highly expressed in the tumor as a mutant, p53 expression in malignant tumors was significantly higher than that in benign tumors (P <0.05). (4) The positive rates of p53 in malignant tumors were significantly higher than those in benign tumors when HSP90α and HSP90β were positive (P <0.01), and the positive rates of HSP90α in malignant tumors were significantly higher than those in benign tumors (P <0.05) . Conclusions (1) The sustained stimulation of HSP90α, HSP90β and mutant p53 are the basis of the development of salivary gland malignant tumor. (2) Both HSP90α and HSP90β can be used as important targets for salivary gland tumor therapy. (3) HSP90α may promote tumor growth and malignant transformation through mutated p53 protein.
