目的通过建立UPLC-ESI-IT-TOF/MS分析方法对芪苓温肾消囊方提取物化学成分进行分析,结合文献报道,初步推测可能的起效成分。方法采用Waters ACQUITY UPLC BEH C18色谱柱(100 mm×2.1 mm,1.7μm);以乙腈-0.1%甲酸水溶液为流动相梯度洗脱;体积流量为0.4 m L/min;ESI源正负离子模式检测。结果通过对照品比对、质谱数据以及参考文献,共指认出来源于黄芪、淫羊藿、苍术、丹参4味药材共36个化学成分,其中包括19个黄酮类成分、11个酚酸类成分、4个三萜皂苷类成分、1个倍半萜苷类及1个生物碱类成分。结论对芪苓温肾消囊方提取物的化学成分进行了较为全面的分析,初步阐明了芪苓温肾消囊方的药效物质基础,对于复方中药化学成分物质基础研究及质量控制具有重要意义。
OBJECTIVE: To establish a UPLC-ESI-IT-TOF / MS method for the analysis of chemical constituents in the extract of Tiling and Wenshenxian Capsules. Combined with the literature reports, the possible active ingredients were preliminarily estimated. Methods Waters ACQUITY UPLC BEH C18 column (100 mm × 2.1 mm, 1.7 μm) was used. The mobile phase was eluted with acetonitrile-0.1% formic acid as mobile phase. The volume flow rate was 0.4 m L / min. Results A total of 36 chemical constituents from Astragalus, Epimedium, Atractylodes, Radix Salviae Miltiorrhizae and four kinds of herbs of Radix Salviae Miltiorrhizae were identified through comparison of reference materials, mass spectrometry data and references, including 19 flavonoids and 11 phenolic acids , Four triterpene saponins, one sesquiterpene glycoside and one alkaloid. Conclusion The chemical composition of the extract of Tiling Wenshenxiban a more comprehensive analysis of the initial clarify the pharmacological effects of Qiling Wenshenxiban basis for the chemical composition of the compound Chinese medicine is essential for the quality of basic research and quality control significance.
