目的探讨E-cadherin在环磷酰胺处理后膀胱黏膜中的作用。方法 SD大鼠12只,随机分为处理组和对照组,处理组大鼠给与环磷酰胺100mg·kg~(-1)腹腔注射,对照组给与同等剂量的生理盐水腹腔注射,24h后取膀胱,观察形态变化,检测E-cadherin mRNA表达水平、表达量。结果肉眼观处理组膀胱出现明显的水肿、出血、粘连;HE染色可见,膀胱黏膜弯曲减少、肿胀,上皮细胞层次减少,上皮变薄,膀胱黏膜呈现毛刺样改变;免疫组化可见,E-cadherin只表达在膀胱黏膜,处理组表达低于对照组;Real-time PCR与Western blot结果可见,E-cadherin mRNA与蛋白的表达量处理组均低于对照组(P<0.05)。结论大鼠腹腔注射环磷酰胺致上皮细胞严重损伤,E-cadherin在膀胱黏膜中表达下调。
Objective To investigate the role of E-cadherin in bladder mucosa after cyclophosphamide treatment. Methods Twelve SD rats were randomly divided into treatment group and control group. The rats in the treatment group were given intraperitoneal injection of cyclophosphamide (100 mg · kg -1) and the control group were injected intraperitoneally with the same dose of saline. After 24 h Bladder was taken for observation of morphological changes. The expression of E-cadherin mRNA was detected. Results Edema, hemorrhage and adhesions were observed in the bladder of the macroscopic observation group. HE staining showed that the bowel mucosa of the bladder mucosa was reduced in curvature, swelling and epithelial cells were reduced, the epithelium was thinner, and the mucosa of the bladder showed burr-like changes. E-cadherin The results of Real-time PCR and Western blot showed that the expressions of E-cadherin mRNA and protein were all lower than those of the control group (P <0.05). Conclusion Cyclophosphamide-induced epithelial cells in rats are severely damaged and E-cadherin is down-regulated in bladder mucosa.
