目的 研究树突状细胞 (DC)体外诱导的抗肿瘤免疫能否抑制裸鼠移植瘤生长并预防其发生。方法 联合应用粒 /巨噬细胞集落刺激因子 (GM -CSF)及白介素 4(IL 4)直接从胃癌患者外周血中培养出DC ;以SGC -790 1细胞的肿瘤抗原粗提物刺激DC使其激活同源的T淋巴细胞产生细胞毒性T淋巴细胞 (CTL) ;建立裸鼠SGC -790 1细胞移植瘤模型 ;DC诱导的CTL治疗裸鼠SGC -790 1细胞移植瘤 ,观察移植瘤生长 ;DC诱导的CTL预防性治疗裸鼠 ,观察随后接种的SGC -790 1细胞移植瘤发生情况。结果 DC诱导的CTL不仅能抑制裸鼠移植瘤生长并能预防其发生。结论 经肿瘤抗原激活的DC作为一新概念上的抗肿瘤疫苗有可能在治疗肿瘤及预防其术后复发和转移中发挥重要作用。
Objective To investigate whether anti-tumor immunity induced by dendritic cells (DCs) in vitro can inhibit the growth of transplanted tumors in nude mice and prevent their occurrence. Methods DCs were cultured directly from the peripheral blood of gastric cancer patients using granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin 4 (IL 4). DCs were stimulated with crude extracts of tumor antigens from SGC-7901 cells. Activation of homologous T lymphocytes to produce cytotoxic T lymphocytes (CTL); Establishment of a nude mouse model of SGC-790 1 cell xenografts; DC-induced CTL treatment of nude mice SGC790 cell xenografts; observe the growth of xenografts; Induction of CTL prophylactically treats nude mice and observation of the subsequent occurrence of transplanted SGC-790 1 cell xenografts. Results DC-induced CTL not only inhibited the growth of transplanted tumor in nude mice but also prevented its occurrence. Conclusion The tumor-activated DCs as a new conceptual anti-tumor vaccine may play an important role in the treatment of tumors and their prevention of postoperative recurrence and metastasis.
