目的:观察思的明(SDM)对纳洛酮催促下的吗啡戒断大鼠不同脑区谷氨酸(Glu)含量的影响。方法:以剂量递增法连续皮下注射(sc)吗啡,建立吗啡依赖大鼠模型,取纳洛酮催促戒断后各组大鼠的皮层和海马。采用2,4-二硝基氟苯(DN-FB)柱前衍生高效液相色谱法(HPLC)测定皮层和海马中Glu的含量。结果:与生理盐水(NS)组比较,SDM对照组皮层和海马中Glu含量差异无显著性,模型组大鼠皮层和海马内Glu的含量明显增高。与模型组比较,SDM治疗组、阳性药可乐定组大鼠皮层和海马中Glu的含量均明显降低。结论:SDM能调节中枢的Glu含量,这可能与SDM可显著抑制大鼠吗啡戒断症状有关。
OBJECTIVE: To observe the effect of simethasin (SDM) on glutamate (Glu) content in different brain regions of morphine withdrawal rats induced by naloxone. METHODS: Morphine-dependent morphine was established by subcutaneous injection of morphine (sc) in a dose-escalating manner, and naloxone was used to induce cortical and hippocampal formation in rats after cessation. The content of Glu in cortex and hippocampus was determined by high performance liquid chromatography (HPLC) using 2,4-dinitrofluorobenzene (DN-FB) precolumn derivatization. RESULTS: Compared with the saline (NS) group, there was no significant difference in Glu content between the cortex and hippocampus of the SDM control group. The content of Glu in the cortex and hippocampus of the model group increased significantly. Compared with the model group, the content of Glu in the cortex and hippocampus of the SDM-treated group and the positive drug clonidine group was significantly reduced. Conclusion: SDM can regulate the central Glu content, which may be related to SDM can significantly inhibit the morphine withdrawal symptoms in rats.
