目的探讨富血小板血浆(platelet-rich plasma,PRP)对体外培养的根尖乳头干细胞(stem cells fromapical papilla,SCAP)生物学效应的影响。方法将SCAP细胞分别在标准培养液、PRP培养液、矿化诱导液及PRP+矿化诱导液中培养。通过缩胆囊素八肽(cell counting kit-8,CCK-8)法、碱性磷酸酶活性(alkaline phosphatase activity,ALP)检测、Western Blotting检测SCAP增殖及分化能力的改变。结果 PRP组及PRP+矿化诱导液组SCAP的增殖能力强于其他组(P<0.05)。与其他组相比较,PRP+矿化诱导液组SCAP的ALP活性及牙本质涎蛋白(dentin sialophosprotein,DSP)表达水平明显上调,矿化能力显著提高(P<0.05)。结论 PRP可明显促进SCAP细胞的增殖能力,与矿化诱导液联合应用对SCAP的矿化能力具有促进作用。
Objective To investigate the effect of platelet-rich plasma (PRP) on the biological effects of stem cell fromapical papilla (SCAP) cultured in vitro. Methods SCAP cells were cultured in standard medium, PRP medium, mineralization inducing fluid and PRP + mineralizing inducing fluid respectively. The proliferation and differentiation of SCAP were detected by cell counting kit-8 (CCK-8) assay, alkaline phosphatase activity (ALP) assay and Western Blotting. Results The proliferative ability of SCAP in PRP group and PRP + mineralization inducing fluid group was stronger than that in other groups (P <0.05). Compared with other groups, the ALP activity and the expression of dentin sialophosprotein (DSP) in SCAP of PRP + mineralization inducing fluid group were significantly up-regulated and the mineralization ability was significantly increased (P <0.05). Conclusion PRP can significantly promote the proliferation of SCAP cells, and mineralization inducing fluid combined with SCAP mineralization has a promoting effect.
