目的研究p62基因在黑素瘤A375细胞生长增殖及细胞周期调控等方面的作用。方法取对数生长期的A375细胞,用干扰p62基因表达的siRNA慢病毒载体转染细胞;MTT法检测转染及正常A375细胞的平均OD值;流式细胞术检测两组的细胞周期分布。结果转染组的平均OD值较正常组小(P<0.05);转染组的G0/G1期比例(0.73±0.01)较正常组(0.64±0.02)高(P=0.002);转染组的凋亡率(0.10±0.02)较正常组(0.03±0.01)高(P=0.004)。结论干扰p62表达后,细胞周期阻滞于G0/G1期,p62在维持细胞周期和肿瘤细胞凋亡中起着重要的作用。
Objective To study the role of p62 gene in the proliferation and cell cycle regulation of melanoma A375 cells. Methods A375 cells in logarithmic growth phase were transfected with siRNA lentiviral vector which interfered with the expression of p62 gene. The mean OD value of transfected and normal A375 cells was determined by MTT assay. The cell cycle distribution was analyzed by flow cytometry. Results The average OD value in the transfected group was significantly lower than that in the normal group (P <0.05). The G0 / G1 phase ratio (0.73 ± 0.01) in the transfected group was significantly higher than that in the normal group (0.64 ± 0.02) (0.10 ± 0.02) was higher than that of normal group (0.03 ± 0.01) (P = 0.004). Conclusions After p62 expression is blocked, the cell cycle arrests at G0 / G1 phase and p62 plays an important role in maintaining cell cycle and tumor cell apoptosis.
