目的探讨核转录因子(NF)-κB p65的阻断剂SN50对裸鼠人胃癌移植瘤生长的影响。方法建立裸鼠胃癌移植瘤模型后将20只裸鼠随机分成对照组(n=5)和SN50实验组(n=15),用生理盐水处理对照组、SN50处理实验组,于第5 d、10 d、15 d分别取5只裸鼠测量肿瘤体积,计算抑瘤率。采用免疫组织化学染色观察凋亡指标Bcl-2、Bax和p53表达的变化。结果使用SN50处理肿瘤后5 d、10 d、15 d肿瘤生长抑制率逐渐增高;与对照组相比,凋亡指标Bcl-2的表达下调,p53和Bax的表达上调(均P<0.05)。结论 NF-κB p65的阻断剂SN50可能通过下调Bcl-2的表达和上调p53、Bax的表达导致细胞凋亡,从而最终抑制移植瘤的生长。
Objective To investigate the effect of SN50, a blocker of NF-κB p65, on the growth of human gastric cancer xenografts in nude mice. Methods Twenty nude mice were randomly divided into control group (n = 5) and SN50 experimental group (n = 15). The control group was treated with saline and the SN50 treatment group. On day 5, On day 10 and day 15, tumor volume was measured in 5 nude mice, and the tumor inhibition rate was calculated. The changes of apoptosis index Bcl-2, Bax and p53 expression were observed by immunohistochemical staining. Results Compared with the control group, the tumor growth inhibitory rate of SN50 increased gradually at 5 d, 10 d and 15 d after treatment with SN50. The expression of Bcl-2 and p53 and Bax were up-regulated compared with the control group (all P <0.05). Conclusion SN50, a blocker of NF-κB p65, may ultimately inhibit the growth of xenografts by down-regulating the expression of Bcl-2 and up-regulating the expression of p53 and Bax.
