目的研究糖尿病视网膜病变患者血清中miRNA-29的表达水平,分析并探讨miRNA-29在糖尿病视网膜病变发生机制中的作用。方法选取健康受检者、糖尿病患者及糖尿病伴有视网膜病变患者,对所参加实验者静脉取血,提取并纯化血浆总RNA,并通过逆转录聚合酶链反应等实验技术获取并检测miRNA-29的表达水平。结果糖尿病视网膜病变组患者和糖尿病不伴并发症患者相比miRNA-29的表达显著增高,差异具有统计学意义(P<0.01),无并发症的糖尿病患者组和健康对照组比较,miRNA-29的表达无明显改变,差异无统计学意义(P>0.05);糖尿病视网膜病变患者组和不伴并发症患者糖尿病患者组、健康对照组相比,RAX蛋白表达水平明显降低,差异具有统计学意义(P<0.05)结论miRNA-29在糖尿病视网膜病变患者血清中表达明显上调,miRNA-29潜在靶基因PKR结合蛋白X(RAX)表达增高,其可能参与糖尿病视网膜病变的发生机制,进而引起糖尿病视网膜病变。
Objective To study the expression of miRNA-29 in serum of patients with diabetic retinopathy and to explore the role of miRNA-29 in the pathogenesis of diabetic retinopathy. Methods Healthy subjects, diabetic patients and patients with diabetic retinopathy were enrolled. Blood samples were taken from venous blood of experimental subjects, total RNA was extracted and purified, and miRNA-29 was obtained and tested by reverse transcription-polymerase chain reaction The level of expression. Results The expression of miRNA-29 in diabetic retinopathy group was significantly higher than that in non-diabetic diabetic group (P <0.01). Compared with healthy controls, miRNA-29 (P> 0.05).
