目的研究红树植物拟海桑Sonneratia paracaseolaris的化学成分。方法利用硅胶柱层析、凝胶Sephadex LH-20柱层析、HPLC等手段对化学成分进行了分离纯化;通过理化性质,结合波谱数据分析并与文献对照,鉴定化合物的结构,分别以MTT和SRB法评价化合物的抗肿瘤活性。结果从红树植物拟海桑Sonneratia paracaseolaris的甲醇提取物中,分离鉴定了9个单体化合物:9-氧橙花叔酮(1),3,7,11-三甲基-1,7(E),10-十二碳三烯-3-羟基-9-酮(2),(22E,24R)-5α,8α-过氧化麦角甾-23-甲基-6,22-二烯-3β-醇(3),(22E)-5α,8α-过氧化麦角甾-23-甲基-6,9,22-三烯-3β-醇(4),(22E)-5α,8α-过氧化麦角甾-6,22-二烯-3β-醇(5),4′-甲氧基偏柏脂素(6),偏柏脂素(7),3′-羟基-4′-甲氧基-4′-脱羟基偏柏脂素(8),1,3-二对羟基苯基-4-戊烯-1-酮(9)。化合物6~9对A549、HL-60、P388及K562肿瘤细胞株显示出不同程度的抑制作用,其中化合物8对K562、HL-60细胞具有较强的细胞毒活性,IC50值分别为1.99、3.13μmol/L。结论化合物1~9均为首次从拟海桑Sonneratia paracaseolaris中分离得到;首次测定了化合物6~9对A549、HL-60、P388及K562肿瘤细胞株的细胞毒活性,其中化合物8对HL-60、K562细胞显示出较好的抑制活性。
Objective To study the chemical constituents of Sonneratia paracaseolaris, a mangrove plant. Methods The chemical constituents were separated and purified by silica gel column chromatography, Sephadex LH-20 column chromatography and HPLC. The structures of the compounds were identified by physical and chemical properties, spectroscopic data analysis and literature comparison. MTT and The anti-tumor activity of compounds was evaluated by SRB method. Results Nine monomer compounds were isolated from the methanol extract of Sonneratia paracaseolaris, a mangrove plant: 9-oxalicuminone (1), 3,7,11-trimethyl-1,7 E), 10-dodecatriene-3-hydroxy-9-one (2), (22E, 24R) -5α, 8α-ergosta-2,3- (4), (22E) -5α, 8α-peroxidation of (22E) -5α, 8α-ergosta-2,3- (5), 4’-methyloxyparamin (6), procaponin (7), 3’-hydroxy-4’-methoxy -4’-dehydroxyquinazolin (8), 1,3-diphenyloxyphenyl-4-penten-1-one (9). Compounds 6 to 9 showed different degrees of inhibition on A549, HL-60, P388 and K562 tumor cell lines, of which Compound 8 had strong cytotoxic activity on K562 and HL-60 cells with IC50 values of 1.99 and 3.13 μmol / L. Conclusions Compounds 1 to 9 were isolated from Sonneratia paracaseolaris for the first time. The cytotoxic activities of compounds 6 to 9 against A549, HL-60, P388 and K562 tumor cell lines were determined for the first time. , K562 cells showed better inhibitory activity.
