刺五加总苷对REM睡眠剥夺所致大鼠记忆巩固障碍及海马群峰电位的影响

目的:研究刺五加总苷对快动眼睡眠剥夺(REM SD)所致记忆巩固障碍的改善作用及对高频刺激(HFS)前后海马群峰电位(PS)幅值的影响。方法:利用六角迷宫对成年雄性SD大鼠进行训练,将确定学会任务的大鼠随机分为大平台对照组(CON)、模型组(SD)和刺五加总苷50,100和200 mg·kg~(-1)3个剂量组。各组大鼠分别进行记忆巩固训练,连续训练4 d。每天训练前30 min灌胃给药或蒸馏水,训练后立即将大鼠放入睡眠剥夺箱进行6 h的REM睡眠剥夺;巩固训练结束后进行测试并利用电生理学方法,记录海马齿状回颗粒细胞层高频刺激HFS前后PS幅值的变化。结果:与CON组相比,SD组大鼠的记忆能力降低(寻找时间延长,错误次数增多,认知率降低),同时海马脑区HFS后PS幅值没有明显增大;与SD组大鼠相比,刺五加总苷50,100及200 mg·kg~(-1)均能明显增强SD大鼠的记忆能力(寻找时间缩短,错误次数减少,认知率提高),并明显升高HFS后PS幅值,具有剂量依赖性。结论:REM睡眠剥夺能够造成大鼠记忆巩固障碍,刺五加总苷能够改善这种障碍,此作用可能与增强大鼠海马区突触可塑性有关。 Objective: To study the effects of acanthopanax senticosus glycosides on improving memory consolidation disorder induced by REM SD and the amplitude of hippocampal peak potentials (PSs) before and after high-frequency stimulation (HFS). Methods: Adult male Sprague-Dawley rats were trained by hexagonal maze. The rats whose task of learning were determined were randomly divided into large platform control group (CON), model group (SD) and Acanthopanax senticosus glycosides 50, 100 and 200 mg · kg ~ (-1) -1) 3 dose groups. Each group of rats were memory consolidation training, continuous training 4 d. Rats were administered intragastrically or distilled water 30 minutes prior to training, and immediately after training, rats were placed in sleep deprivation box for 6-h REM sleep deprivation. After consolidation training, the rats were tested and electrophysiological methods were used to record the changes of hippocampal dentate gyrus granulosa cells Changes of PS amplitude before and after HFS stimulation. Results: Compared with CON group, the memory ability of rats in SD group decreased (search time increased, error times increased and cognitive rate decreased), while the amplitude of PS in hippocampus did not increase significantly after HFS. Compared with CON group, Compared with the control group, acanthopanax senticosus glycosides 50, 100 and 200 mg · kg -1 could significantly improve the memory ability of SD rats (shortened the search time, reduced the number of errors, and improved the cognition rate) and significantly increased the HFS PS amplitude, in a dose-dependent manner. Conclusion: REM sleep deprivation can cause impairment of memory consolidation in rats, and acanthopanax senticosus can improve this disorder, which may be related to the enhancement of synaptic plasticity in rat hippocampus.
