目的:探讨多器官功能不全(MODS)小鼠在伤后不同时相点白介素-12(IL-12)水平的变化趋势及其与创伤后免疫失衡的关系。方法:选择6~8周龄雄性 C57BL/6小鼠96只,随机分为正常组,伤后3~24h 组、3~7d 组和12d 组。腹腔注射酵母多糖复制 MODS 动物模型,用间接酶标法行脾脏 IL-12p40的免疫组织化学标记,采用 ELISA 法分别检测血清与脾脏组织匀浆上清中 IL-12p40和 IL-12p70的水平,流式细胞术检测外周血 T细胞亚群。结果:在 MODS 病程早期和晚期的“两次打击”中,小鼠脾脏组织与血清中 IL-12p40含量显著增多,IL-12p70相对减少,同时伴有 CD4/CD8比值下降。结论:MODS 小鼠发生免疫抑制可能与免疫器官中抗原提呈细胞高表达 IL-12p40有一定相关性。
Objective: To investigate the change trend of interleukin-12 (IL-12) and its relationship with posttraumatic immune imbalance in multiple organ dysfunction (MODS) mice at different time points after injury. Methods: Ninety-six male C57BL / 6 mice aged 6-8 weeks were randomly divided into normal group, 3 ~ 24h group, 3 ~ 7d group and 12d group. The animal model of MODS was induced by intraperitoneal injection of zymosan, and the spleen IL-12p40 was detected by indirect enzyme-linked immunosorbent assay. The levels of IL-12p40 and IL-12p70 in serum and spleen homogenate supernatants were measured by ELISA Cytometry was used to detect peripheral blood T cell subsets. Results: The levels of IL-12p40 in spleen tissues and serum of mice were significantly increased and IL-12p70 was relatively decreased with the decrease of CD4 / CD8 ratio in early and late stages of MODS. Conclusion: The immunosuppression in MODS mice may be related to the high expression of IL-12p40 in antigen presenting cells in immune organs.
