观察软肝化疒徵丸干预肝纤维化的主要作用机制。分离培养大鼠肝星状细胞 (HSC) ,以软肝化疒徵 丸灌胃正常大鼠 ,制备药物血清 ,温育培养细胞。 3H TdR掺入法测定细胞增殖 ,ELISA法测定细胞上清Ⅰ型胶原含量 ,RT PCR法观察前胶原α2 (Ⅰ )mRNA的表达 ,同位素标记法测定细胞上清间质胶原酶的活性。结果 :软肝化疒徵丸药物血清能抑制HSC的增殖与胶原蛋白的分泌 ,提高其间质胶原酶的活性 ,抑制其α2 (Ⅰ )mRNA的表达。结论 :软肝化疒徵 丸干预肝纤维化的作用机制可能在于抑制HSC的活化
Observe the main mechanism of intervention of Ruangan Huayu Pill on liver fibrosis. Rat hepatic stellate cells (HSC) were isolated and cultured, and normal rats were given intragastric administration of Ruangan Huayu Zhengqi pills, and drug serum was prepared and incubated to cultivate cells. 3H TdR incorporation method was used to determine cell proliferation. ELISA was used to determine the content of type I collagen in the cell supernatant. RT-PCR was used to observe the expression of procollagen α2 (I) mRNA. The activity of collagenase in the supernatant was determined by isotope labeling. Results : The drug serum of Ruangan Huayu Quzhen pill could inhibit the proliferation of HSC and secretion of collagen, increase the activity of interstitial collagenase and inhibit the expression of α2(I) mRNA. Conclusion : The mechanism of intervention of liver stagnation and phlegm granules on liver fibrosis may be to inhibit the activation of HSC
