目的检测Cath-D和MMP-9和在皮肤恶性黑色素瘤、交界痣及正常皮肤中表达情况,及其与皮肤恶性黑色素瘤的临床分期,淋巴结转移的关系。方法应用免疫组化S-P法对分别检测Cath-D和MMP-9在44例皮肤恶性黑色素瘤、22例皮肤交界痣及20例正常皮肤中的表达水平。结果 Cath-D和MMP-9在皮肤恶性黑色素瘤中的阳性表达率(79.55%和75.00%)均高于交界痣(45.45%和40.91%)及正常皮肤组织(15.00%和30.00%)(P<0.05),Cath-D和MMP-9在皮肤恶性黑色素瘤中的阳性表达率随肿瘤浸润深度的增加而增高,且有淋巴结转移者阳性表达率大于无淋巴结转移者(P<0.05)。皮肤恶性黑色素瘤中Cath-D和MMP-9的表达呈正相关(rs=0.488,P<0.05)。结论 Cath-D和MMP-9在皮肤恶性黑色素瘤的发生、发展中可能起着重要作用,且与CMM的浸润转移密切相关,且二者之间可能具有协同作用。联合检测两者的表达情况可为CMM的早期诊断及预后提供新的靶点。
Objective To detect the expression of Cath-D and MMP-9 in cutaneous malignant melanoma, border nevus and normal skin and their relationship with clinical stage and lymph node metastasis of cutaneous malignant melanoma. Methods Immunohistochemical S-P method was used to detect the expression of Cath-D and MMP-9 in 44 cases of cutaneous malignant melanoma, 22 cases of cutaneous border mole and 20 cases of normal skin. Results The positive rates of Cath-D and MMP-9 in skin malignant melanoma (79.55% and 75.00%) were significantly higher than those in border nevi (45.45% and 40.91%) and normal skin tissues (15.00% and 30.00% <0.05). The positive expression rate of Cath-D and MMP-9 in the skin malignant melanoma increased with the depth of tumor invasion, and the positive expression rate of Cath-D and MMP-9 was higher than that without lymph node metastasis (P <0.05). There was a positive correlation between the expression of Cath-D and MMP-9 in skin malignant melanoma (rs = 0.488, P <0.05). Conclusions Cath-D and MMP-9 may play an important role in the occurrence and development of cutaneous malignant melanoma, and are closely related to the invasion and metastasis of CMM, and may play a synergistic role between them. Combined detection of the expression of the two may provide a new target for the early diagnosis and prognosis of CMM.
