苯丙氨酸解氨酶(phenylalanine ammonialyase,PAL)是酚类物质合成的第一个酶,是催化苯丙烷类反应的第一酶。根据已经报道的PAL基因的序列设计兼并引物,采用3′RACE和5′RACE方法,克隆得到了芒果果实PAL基因的全长cDNA序列。该基因开放阅读框为2160 bp,编码719个氨基酸,分子重量为79.18 kD。对基因组扩增得到了2200 bp长度的片段分析发现,该基因未含有内含子序列。通过系统发育分析发现,该基因编码的蛋白与问荆、杉木、银杏等植物具有较近的亲缘关系。对不同芒果品种的PAL基因的表达进行分析发现,红色的贵妃品种中表达量较高,而绿色的桂七品种中表达量较低。
Phenylalanine ammonialyase (PAL) is the first enzyme that synthesizes phenolic compounds and is the first enzyme to catalyze the reaction of phenylpropanes. According to the published sequence of PAL gene, the full length cDNA of PAL gene was cloned by 3’RACE and 5’RACE. The open reading frame of this gene was 2160 bp, encoding 719 amino acids with a molecular weight of 79.18 kD. A fragment of 2200 bp was amplified from the genome and found that the gene did not contain an intron sequence. Phylogenetic analysis showed that the protein encoded by this gene has a close genetic relationship with plants such as Zing Jing, Chinese fir and Ginkgo biloba. The PAL gene expression of different mango cultivars was analyzed. The results showed that the expression of PAL gene in red cabbage was higher, while that in green Guiqi was lower.
