采用四氯化碳(CCl4)制备小鼠肝纤维化模型,检测AST,ALT,γ-GT,MDA,GSH-px,SOD等血清指标,ELISA方法检测PCⅢ,IV-C,LN,HA等血清标记物;肝组织采用HE和Masson胶原染色,光镜下观察病理学改变;Western blot检测Ⅲ型胶原、TGF-β,α-SMA,E-cadherin蛋白表达,观察虎眼万年青提取物对CCl_4诱导的小鼠肝纤维化治疗作用,并探讨作用机制。结果表明,虎眼万年青提取物组(50,150,500 mg·kg~(-1))均可明显降低血清AST,ALT,γ-GT,MDA水平,升高GSH-px,SOD水平;降低肝纤维化血清标志物PCⅢ,IV-C,LN,HA的表达;明显改善CCl_4所致大鼠肝纤维化的病理学改变,证明虎眼万年青提取物对肝纤维化具有治疗作用。其机制是可能与抑制细胞外基质沉积,抑制肝星形细胞活化以及抑制上皮间质转化,进而抑制肝脏纤维化有关。
Serum levels of AST, ALT, γ-GT, MDA, GSH-px and SOD were detected by carbon tetrachloride (CCl4), serum of PCⅢ, IV-C, LN and HA were detected by ELISA The hepatic tissues were stained with HE and Masson collagen, and the pathological changes were observed under light microscope. The expressions of type Ⅲ collagen, TGF-β, α-SMA and E-cadherin were detected by Western blotting. Mice liver fibrosis treatment, and explore the mechanism of action. The results showed that the extract of Ornithogalum album could significantly reduce the level of serum AST, ALT, γ-GT and MDA, and increase the levels of GSH-px and SOD, and reduce the serum levels of hepatic fibrosis The expressions of PCⅢ, IV-C, LN and HA were significantly improved. The pathological changes of hepatic fibrosis induced by CCl_4 were significantly improved, which indicated that the extract of Ornithogalum Cymbidium had a therapeutic effect on hepatic fibrosis. The mechanism may be related to the inhibition of extracellular matrix deposition, inhibition of hepatic stellate cell activation and inhibition of epithelial-mesenchymal transition, thereby inhibiting hepatic fibrosis.
