目的 :探讨端粒酶活性及端粒酶hTRT表达在胃癌发生及发展中的作用。方法 :分别采用TRAP银染法和RT PCR法对胃粘膜端粒酶活性及端粒酶hTRTmRNA进行检测。结果 :胃癌组织端粒酶阳性率为 1 0 0 % (8/8) ,胃癌前状态组织端粒酶阳性率为 40 % (4/1 0 ) ,浅表性胃炎均为阴性 (0 /9)。胃癌组织hTRTmRNA阳性率为 85 .7% (6/7) ,浅表性胃炎均为阴性 (0 /5 )。结论 :端粒酶激活参与胃癌的形成和发展 ,可能有助于胃癌的早期诊断。胃粘膜端粒酶hTRT表达水平可能反映端粒酶存在和活化的程度。
Objective: To investigate the role of telomerase activity and telomerase hTRT expression in the development and progression of gastric cancer. Methods: The gastric mucosal telomerase activity and telomerase hTRT mRNA were detected by TRAP silver staining and RT PCR respectively. Results: The positive rate of telomerase was 100% (8/8) in gastric cancer tissues, 40% (4/1 0) in gastric precancerous lesions, and 0/9 ). The positive rate of hTRT mRNA in gastric cancer tissues was 85.7% (6/7) and the superficial gastritis was all negative (0/5). Conclusion: The involvement of telomerase activation in the formation and development of gastric cancer may contribute to the early diagnosis of gastric cancer. The level of telomerase hTRT in gastric mucosa may reflect the presence and activation of telomerase.
