目的运用代谢组学方法研究脾气虚证大鼠尿液的代谢表型变化,筛选出与脾气虚证相关的代谢标志物,并探讨补中益气汤的治疗作用。方法利用核磁共振氢谱(1H-NMR)技术建立大鼠尿液的代谢指纹谱。应用主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)研究正常对照组与模型组、模型组与补中益气汤组之间的代谢物谱差异,并通过变量重要性投影(VIP)在尿液中选取生物标志物。结果模型组及补中益气汤组的代谢表型发生了明显的变化,并筛选出与脾气虚证相关的9种代谢产物,补中益气汤对部分生物标志物有逆转作用。结论脾气虚证大鼠的脂质及氨基酸代谢都出现了紊乱,能量代谢降低;经补中益气汤治疗后大鼠蛋白质代谢明显增强,保证了大鼠机体能量的供给。
OBJECTIVE: To study the metabolic phenotype of urine in rats with spleen-qi deficiency syndrome by using metabonomics, and to screen out the metabolic markers related to spleen deficiency syndrome and to explore the therapeutic effect of Buzhong Yiqi Decoction. Methods The metabolic fingerprint of rat urine was established by 1H-NMR. Principal Component Analysis (PCA) and Orthogonal Partial Least Squares Discriminant Analysis (OPLS-DA) were used to study the difference of metabolites profiles between normal control group and model group, model group and Buzhongyiqi Decoction group, Sexual Projection (VIP) Select biomarkers in urine. Results The metabolic phenotypes of the model group and Buzhong Yiqi Decoction group were significantly changed, and nine metabolites related to the spleen qi deficiency syndrome were screened out. Buzhong Yiqi Decoction reversed some of the biomarkers. Conclusion The lipid metabolism and amino acid metabolism of rats with spleen-qi deficiency syndrome were disordered and the energy metabolism was decreased. The protein metabolism of rats was obviously enhanced after treated by Buzhong Yiqi Decoction, which ensured the energy supply of rats.
