报道了酚-次氯酸盐直接显色法测定血氨。结果表明,显色反应在pH11.0~11.6范围内吸光度大、稳定且干扰小。有色化合物最大吸收波长位于635nm,表观摩尔吸光系数为1.83×104L·mol-1·cm-1,血氨浓度在0~1200μmol/L范围内符合比耳定律,检测下限为4.8μmol/L。采用两种碱浓度不同的次氯酸盐溶液分别测定标准和血液样品,保持了显色溶液酸度一致性,因而提高了血氨测定的准确度,回收率达98.5%~101.3%。本方法对35例正常人和22例高血氨患者进行血氨含量测定,结果均令人满意。本方法可在临床推广应用。
Reported the phenol - hypochlorite direct colorimetric determination of ammonia. The results showed that the color reaction in the pH 11.0 ~ 11.6 absorbance range, stable and small interference. The maximum absorption wavelength of the colored compound is at 635nm, the apparent molar absorptivity is 1.83 × 104L · mol-1 · cm-1, and the blood ammonia concentration is in the range of 0-1200μmol / L. The lower limit of detection is 4.8μmol / L. Hypochlorite solutions with different alkali concentrations were used to determine the standard and blood samples separately and maintain the acidity consistency of the chromogenic solution, thus improving the accuracy of the blood ammonia determination with a recovery of 98.5% -101.3% . The method of 35 cases of normal and 22 cases of hypernatremia in patients with serum ammonia determination, the results were satisfactory. The method can be applied in clinic.
