【目的】研究OT型百合品种黄天霸花器官的离体培养快速繁殖技术,为其种球产业化生产提供技术支持。【方法】以黄天霸花蕾和半开花的不同部位为外植体,以MS为基本培养基,探讨不同外植体的愈伤组织诱导效果、0~3.0 mg/L6-BA对外植体愈伤组织分化、1.0~3.0 mg/L 6-BA+0~0.2 mg/L NAA组合对叶片不定芽诱导、30.0~80.0 mg/L蔗糖对鳞茎形成及生根的影响,调查不同基质对组培苗移栽种植的效果。【结果】花蕾和半开花不同部位愈伤组织诱导从易到难表现为:子房>柱头>花药>花托>花丝、花瓣,子房和柱头的愈伤组织诱导率均超过50.0%。在MS培养基中,随着6-BA用量的增加(0~3.0 mg/L),外植体愈伤组织分化率先提高后稍有下降,适宜分化培养基为MS+2.5 mg/L 6-BA。在不同6-BA和NAA组合中,以培养基MS+3.0 mg/L 6-BA的无菌叶片不定芽诱导效果较优。添加60.0 g/L蔗糖的MS培养基利于不定芽结鳞茎生根;带鳞茎组培苗移栽于大田菜园土即可成活。【结论】成功建立的OT型百合品种黄天霸花器官离体快速繁殖体系可用于其种球工厂化生产,扩大规模化种植。
【Objective】 The purpose of this study was to study the rapid propagation technology of organotypic growth in vitro of yellow lily bulbs of OT type lily and to provide technical support for the industrialization of bulblets. 【Method】 The explants were treated with different parts of flower buds and semi-flowering explants and MS as basic medium to investigate the callus induction effects of different explants. The effects of 0 ~ 3.0 mg / L 6-BA on explants The effects of different combinations of 1.0 ~ 3.0 mg / L 6-BA + 0 ~ 0.2 mg / L NAA on adventitious buds induction, 30.0 ~ 80.0 mg / L sucrose on bulb formation and rooting were investigated. Transplanting planting effect. 【Result】 The results showed that the callus induction in different parts of bud and semi-flowering from easy to difficult were: ovary> stigma> anther> receptacle> filament, petal, ovary and stigma callus induction rate exceeded 50.0%. In MS medium, with the increase of 6-BA (0-3.0 mg / L), the callus differentiation rate of explants first increased slightly and then decreased slightly. The appropriate differentiation medium was MS + 2.5 mg / L 6- BA. In different combinations of 6-BA and NAA, the induction of adventitious buds by sterile leaves of MS + 3.0 mg / L 6-BA was better. MS medium supplemented with 60.0 g / L sucrose was beneficial to shoots of adventitious buds and bulbous shoots; 【Conclusion】 The successful establishment of the OT-type lily variety Huangtianba flower organs in vitro rapid propagation system can be used for its bulbs factory production, expand large-scale cultivation.
