目的观察胫骨癌痛模型大鼠脊髓STAT3的表达,探讨其与星形胶质细胞的关系。方法选择雌性SD大鼠20只,随机分为2组:模型组(n=10),为左侧胫骨上段骨髓腔注入3μlMADB-106大鼠乳腺癌细胞(4.8×109/L);对照组(n=10),为左侧胫骨上段骨髓腔注入3μlHank液。第14天,采用免疫组织化学方法和荧光双标法观察骨癌痛模型大鼠患侧腰段脊髓STAT3和GFAP免疫反应阳性产物的变化和关系。结果左侧胫骨上段骨髓腔注入癌细胞后第14天,骨癌痛模型大鼠患侧脊髓STAT3、GFAP免疫反应阳性产物表达明显增加(P<0.05);荧光双标免疫组化显示患侧脊髓STAT3与星形胶质细胞标志物GFAP有共表达。结论胫骨癌痛敏的产生和维持与脊髓STAT3的表达上调相关,脊髓星形胶质细胞可能通过JAK/STAT3信号通路参与骨癌痛的发生和发展。
Objective To observe the expression of STAT3 in spinal cord of rat model of tibial cancer pain and to explore its relationship with astrocytes. Methods Twenty female Sprague-Dawley rats were randomly divided into 2 groups: model group (n = 10), 3μl MADB-106 rat breast cancer cells (4.8 × 109 / L) n = 10). 3 μl of Hank’s solution was injected into the medullary cavity of the left tibia. On the 14th day, the changes and relationships of STAT3 and GFAP immunoreactive products in the lumbar spinal cord of the bone cancer pain model were observed by immunohistochemistry and fluorescent double labeling. Results The expression of STAT3 and GFAP immunoreactive products in the ipsilateral spinal cord increased significantly (P <0.05) on the 14th day after injection into the left medial tibial bone marrow cavity. Fluorescence double-labeled immunohistochemistry showed that the ipsilateral spinal cord STAT3 is co-expressed with the astrocyte marker GFAP. Conclusions The production and maintenance of pain sensitivity in tibial cancer are related to the upregulation of STAT3 in spinal cord. Spinal cord astrocytes may participate in the occurrence and development of bone cancer pain through JAK / STAT3 signaling pathway.
