目的:研究肾性高血压大鼠局灶性脑缺血再灌注后脑组织Survivin、Bcl- 2表达和细胞凋亡,探讨其意义。方法:体重2 90~3 10g肾性高血压Wistar大鼠5 0只随机分成2组:A组(缺血再灌注组)及B组(假手术组)。采用线栓法制作局灶性脑缺血2h再灌注模型,假手术组不造成缺血。用免疫组化法和TUNEL法分别观察再灌注6、12、2 4、48和72h脑组织Survivin、Bcl- 2表达和凋亡细胞。结果:在缺血再灌注组中:①再灌注6h时较多Survivin表达,到72h仍见强烈表达;②再灌注6h即可见Bcl -2表达较多,2 4h达高峰,此后逐渐下降;③再灌注6h后已出现较多凋亡细胞,在72h达高峰。假手术组及再灌注组的非缺血侧未见Survivin、Bcl- 2阳性细胞,但见0~2个凋亡细胞。结论:局灶性脑缺血再灌注后Survivin、Bcl- 2表达显著增高,细胞凋亡参与了脑缺血再灌注过程。
OBJECTIVE: To investigate the expression of Survivin and Bcl-2 in brain tissue and cell apoptosis after focal cerebral ischemia-reperfusion in renovascular hypertensive rats and its significance. Methods: Fifty male Wistar rats weighing 2 90-3 10 g were randomly divided into two groups: group A (ischemia reperfusion group) and group B (sham operation group). The focal cerebral ischemia 2h reperfusion model was made by the method of thread occlusion, and the sham operation group did not cause ischemia. The expressions of Survivin and Bcl-2 and apoptotic cells were observed by immunohistochemistry and TUNEL at 6, 12, 2, 48 and 72 h after reperfusion. Results: In the ischemia-reperfusion group, the expression of Survivin was more at 6h after reperfusion, and remained strong at 72h; ② The expression of Bcl-2 was more at 6h after reperfusion, then peaked at 24h and then decreased gradually; After 6h of reperfusion, more apoptotic cells appeared and reached the peak at 72h. There were no Survivin and Bcl-2 positive cells in sham-operated group and non-ischemic group in reperfusion group, but 0 ~ 2 apoptotic cells were observed. Conclusion: The expression of Survivin and Bcl-2 is significantly increased after focal cerebral ischemia-reperfusion. Apoptosis is involved in the process of cerebral ischemia-reperfusion.
