目的:探讨醋酸铅对体外培养未成年小鼠骨髓间充质干细胞生长的影响,为进一步阐明铅骨髓毒性作用机制,采取相应防治措施提高儿童健康水平提供依据。方法:不同浓度醋酸铅作用于体外培养的骨髓间充质干细胞,观察其对间充质干细胞成集落(CFU-F)数的影响及变化规律。结果:醋酸铅可明显抑制CFU-F的生长,各实验组与对照组比较差异有统计学意义(P<0.05),且随浓度增加抑制作用增强,呈明显的剂量-效应关系(r=-0.960,P=0.000),其中CFU-F集落的IC50=80.00μmol/L。结论:醋酸铅可明显抑制体外培养的骨髓间充质干细胞生长。
OBJECTIVE: To investigate the effect of lead acetate on the growth of bone marrow mesenchymal stem cells (MMNCs) cultured in vitro in young mice. To provide a basis for further elucidating the mechanism of lead toxicity in bone marrow and taking appropriate prevention and control measures to improve children’s health. Methods: Bone marrow mesenchymal stem cells cultured in vitro were treated with different concentrations of lead acetate, and their effects on the number of CFU-F colonies were observed. Results: Lead acetate significantly inhibited the growth of CFU-F. There was significant difference between the experimental group and the control group (P <0.05), and the inhibitory effect was enhanced with the increase of concentration, showing a dose-response relationship (r = -0.960 , P = 0.000) with IC50 of CFU-F colonies = 80.00 μmol / L. Conclusion: Lead acetate can significantly inhibit the growth of bone marrow mesenchymal stem cells cultured in vitro.
