目的通过检测糖尿病性视网膜病变(DR)患者血浆、眼内液(房水、玻璃体)血管内皮生长因子(VEGF)含量,探讨其在DR发展变化中的作用。方法采用双抗体夹心酶联免疫吸附试验ELISA法定量检测患者组及对照组血浆、房水、玻璃体VEGF含量。结果无糖尿病性视网膜病变(NDR)组血浆VEGF含量(34.47±l.76)pg/mL,单纯型糖尿病性视网膜病变(BDR)组血浆VEGF含量(53.93±3.08)pg/mL,增生型糖尿病性视网膜病变(PDR)组血浆VEGF含量(53.36±3.28)pg/mL,对照组血浆VEGF含量(178.30±10.13)pg/mL,与实验组比较差异均有显著性的意义(P<0.05);PDR组房水VEGF含量(184.86±12.60)pg/mL,对照组房水VEGF含量(90.06±8.32)pg/mL,两者比较差异有显著性的意义(P<0.05);PDR组玻璃体VEGF含量(74l.70±92.02)pg/mL,对照组玻璃体VEGF含量(94.38±21.21)pg/mL,两者比较差异有显著性的意义(P<0.05)。PDR组血浆VEGF与房水、玻璃体VEGF无相关关系(P>0.05),玻璃体VEGF与糖化血红蛋白(HbAlc)有正相关关系(r=0.9067,P<0.01)。结论糖尿病患者血浆VEGF含量较正常人低,但与房水、玻璃体VEGF含量无关;增生型糖尿病性视网膜病变患者房水与玻璃体VEGF含量增高,玻璃体内VEGF含量增高与患者HLAlc值有关。
Objective To investigate the role of vascular endothelial growth factor (VEGF) in the development of DR in patients with diabetic retinopathy (DR). Methods Double antibody sandwich enzyme-linked immunosorbent assay (ELISA) was used to quantitatively detect plasma, aqueous humor and vitreous VEGF levels in patients and controls. Results The plasma levels of VEGF (34.47 ± l.76) pg / mL in non-diabetic retinopathy (NDR) group and 53.93 ± 3.08 pg / mL in patients with simple diabetic retinopathy (BDR) The plasma levels of VEGF (53.36 ± 3.28) pg / mL in the PDR group and 178.30 ± 10.13 pg / mL in the control group were significantly different from those in the experimental group (P <0.05). PDR (P <0.05); VEGF content in PDR group (184.86 ± 12.60) pg / mL and VEGF in control group (90.06 ± 8.32) pg / mL were significantly different 74l.70 ± 92.02) pg / mL, and vitreous VEGF content in the control group (94.38 ± 21.21) pg / mL. There was significant difference between the two groups (P <0.05). Plasma VEGF in PDR group had no correlation with aqueous humor and vitreous VEGF (P> 0.05). There was a positive correlation between vitreous VEGF and HbAlc (r = 0.9067, P <0.01). Conclusions Plasma VEGF levels in diabetic patients are lower than those in normal subjects, but not in aqueous humor and vitreous body. VEGF levels in aqueous humor and vitreous body are increased in patients with proliferative diabetic retinopathy, and VEGF levels in vitreous body are correlated with HLA1c values.
