本文利用羧甲基羟胺(CMO),在睾酮3位引入羧基,经N-羟基琥珀酰亚胺活化羧基后,与异鲁米诺衍生物(ABEI)脱水缩合,合成了睾酮的化学发光标记物(T─3-CMO─ABEI).并对该标记物进行了以下鉴定:(l)mp.128~130℃;(2)硅胶GF-254薄层层析:展开剂为:氯仿:甲醇:氨水=75:30:5,仅有一个斑点Rf=0.86;(3)紫外光谱:λMeHOmax:202,236,284,310nm;(4)快原子轰击质谱:M+l=620该标记物的无水甲醇贮备液,在4℃下可长期稳定,工作试剂72h内未见发光效率下降,发光检测限达0.21pg/管.
